The H-H-H-H-H-H motif is used as a tag on many recombinant proteins to facilitate purification. The antibody recognizes the His-tag fused to the amino- or carboxy- termini of targeted proteins in transfected or transformed cells.

蛋白質純化的新途徑─應用His-Tag抗體檢測His-Tag融合蛋白
1） 6xHis與不同的Sepharose純化填料結合（捆綁），可不受構象的影響，在天然或變形的條件下可進行進一步純化。
2） 可在溫和的洗脫液環境中，其結合、洗滌及洗脫步驟不會對蛋白結構產生影響。
3） 6xHis標簽比普通（Tag）要小得多，經純化后的蛋白可直接進行下游操作。可用于任何表達系統（包括： 細菌，桿狀病毒和哺乳動物）
4） 6xHis標簽在生理溶液pH下不帶電荷，其標簽不會干涉重組蛋白的結構和功能， 6xHis不會影響蛋白分泌。
5） 6xHis標簽免疫原性差，重組蛋白可以不必除去標簽而直接作為抗原進行免疫。 |
